I have the same issue with my S-50.
I have opened it up and checked all components, it looks good, but
looks are deceiving. I cleaned up the phono jacks and sprayed with air
and contact cleaner. No luck.
Someone had mentioned that the components go bad, like the capacitors
or transistors, and would need to be replaced. More work than I want
to invest in the S-50.
If you really want to get it working, there are some web sites where
you can buy replacement circuit boards.
Or just use it as an awesome MIDI controller. :)

I have Roland S-50 sampler in nearly mint condition. Just one minor issue:
periodic overload (overdrive) of output signal. I've tested power/filter
board and it works fine. S-50 is equipped with multivoltage power supply
and I've tried 117 and 220V wirings but always is the same: every 7-8th
notes from S-50 are played distorted. The Visually sampler is in perfect
condition - all seems to work fine, there are no sights of burnt components
inside. This problem exists on all outputs: individual, phones, mix.
